A signal peptide is a short amino acid sequence, typically 16–30 residues long, located at the N-terminus of a newly synthesized protein that directs the protein to the secretory pathway for export outside the cell or insertion into a membrane. Acting as a molecular address label, the signal peptide is recognized by cellular machinery that routes the protein to the endoplasmic reticulum, after which the peptide is cleaved and the mature protein proceeds to its destination. In robotics and AI research, signal peptides are relevant in synthetic biology and biohybrid systems, where engineered microorganisms or cell-based components must reliably secrete functional proteins — such as bioluminescent reporters like Metridia luciferase — into the extracellular environment. Accurate prediction and design of signal peptides is also a key task in computational biology and proteomics workflows. Understanding signal peptides matters because it enables precise control over protein localization and secretion, which is essential for biosensor design, living materials, and AI-driven protein engineering pipelines.
