Here, we describe a protocol for automated library preparation for DNA barcode sequencing. The protocol uses a Hamilton NGS-STAR automated liquid handler equipped with an automated thermocycler. This protocol requires an automated liquid handler (Hamilton Robotics, NGS-STAR) equipped with an automated thermocycler.
